Efficacy of B Vitamin Supplementation on Global Cognitive Function in Older Adults

After excluding statistical outliers and single-blind trials, the pooled effect was very small. Because the interventions combined several B vitamins, the result cannot be attributed to B12 alone.

Effects of Vitamin B12 Supplementation on Cognitive Function, Depressive Symptoms, and Fatigue

No benefit was found for cognition or depressive symptoms when overt deficiency was absent. Only one trial measured idiopathic fatigue, so no pooled conclusion was possible.

Oral versus intramuscular administration of vitamin B12 for vitamin B12 deficiency in primary care: the OB12 trial

Oral treatment was non-inferior at 8 weeks, but the prespecified non-inferiority criterion was not met at 52 weeks. Most participants preferred oral treatment.
